"Enzymatic Histochemical Study of the Rat Liver"

Abstract


In a part of attempting to elucidate the me-tabolic functional activities of the liver, especially of the bile ducts, several enzymatic histachem-ical, such as acid phosphatse, alkaline phospha-tase, adenosinetriphosphatase (ATP-ase) , 5-nuc-leotidase, non-specific esterase, reduced nicotin-5 amide adenine dinucleotide (?vADH) diaphorase and succinic dehydrogenase (SD), activity patterns were studied in the variously proliferated bile
ductds of CFA adult rat livers. These conditions were experimentally producd by means of the common duct ligation for 3 weeks and clonorchis sinensis infestation for 2 months, and comparative studies were carried out with normal ones.
1. On the distribution of acid phosphatase: In normal rat livers, a slight degree of enzymatic activities were observed in the epitheliar cyto-plasm of bile ducts.
The pericanalicular enzymatic activities were markedly increased following the common duct ligation for 3 weeks, while slightly increased following the common duct ligation for 3 weeks, while slightly increased following clonorchis sinensis infestation for 3 weeks, while slightly increased following clonorchis sinensis infestation for 2 months. In the latter case, a slightly increased luminal ´surface area enzymatic activit-ies of the relatively larger bile duct epithelium were observed.
2. On the distribution of alkaline phosphatase: The enzymatic activities in the periductal and ductular connective tissue were observed.
Following the common duct ligation for 3 weeks, a slightly increased activities were observed in them. A slightly increased activities in the periductular connective tissue, but of moderate increase in the periductal connective tissue were observed in the case of clonorchis infestation for months.
3. On the distribution of ATP-ase and 5-nucleotidase: Generally, staining reaction of 5-nucleotidase was weaker than that of ATP-ase. In normal rat livers, the bile canaliculi were demonstrated brilliantly. The ATP-ase activity in the periductal connective tissue was more tve~ikly stained than in bile canaliculi, but 5-nucleotidase revealed of reversed reactions.
A slightly decreased ATP-ase activity was observed in bile canaliculi of the ligated common duct, but markedly increased picture was obtained in the periductal connective tissue. On the luminal surface of the ductal epithelium was. observed a slight degree of the enzymatic activity. In the case of clonorchis sinensis infestation, the pictures of canalicular rupturing, distortion and small bud-like outpouching were very prominent, and ATP-ase activity was markedly decreased than in common duct ligation and normal control. J~Iuch increased enzymatic activity was observed on the luminal surface of the duct epithelium. However, 5-nuclectidase activity was same degree under these different conditions.
4. On the distribution of non-specific esterase: In normal rat livers, bile duct epitheliar cytoplasmic activity was observed, and following the common duct ligation a slightly increased activity was observed in both ductal and ductular epithelium. While in the clonorchis sinensis infested cases, the enzymatic activity was much more increased in the ductal epithelium than in ductular epithelium. Morever, accentuated activity was observed in the vicinity of the bile duct luminal surface.
5. On the distribution of NADH diaphorase and SD: Enzymatic activity in the bile duct epitheliar cytoplasm was observed.
A slightly increased activity following the common duct ligation, but moderately increased in clonorchis sinensis infested cases were observed. In tb.e latter case, the enzymatic activity was markedly increased in ductal epithelium than in ductu.lar epithelium, and with association of markedly accentuated feature on the luminal border of the bile duct epithelium.
Physico-pathological mechanisms studied by means of serveral enzymatic histochemical patterns of the rat livers were discussed.
