Abstract


Colorimetric method for hippuric acid (Umberger, D.J. and Fiorese, F.F. : Clinical Chem., 9:91,1963) was applied for the determination of urinary hippuric acid and led to a test applicable for the quantitative estimation. Urinary hippuric acid, determined by the colorimetric method, was found to be about 60∼70% of administered sodium benzoate.
In attempts to separate urinary C^(14)-compounds by thin layer chromatography after administration of sodium benzoate-C^(l4) (300 μmoles/100g body wt.) two major fractons, hippuric acid-C^(l4), and benzoic acid-C^(l4) were identified.
The total urinary C^(l4) was about 90% of test dose and the ratio of hippuric acid-C^(l4) to benzoic acid-C^(l4) was about 3:1. The ratio was significantly reduced with increased excretion of free benzoic acid in fasting rats. Discussion was made on the advantage of the ratio determination as a more valuable criterion of benzoic acid detoxication than determination of urinary hippuric acid alone in rats.
