Effects of Histamine and Histanine with Chlorpheniramine and Furosemide on Renal function in Anesthetized dogs.

Abstract


Effects of histamine and influence of chlorpheniramine and of chlorpheniramine with furosemide upon the effects of histamine on renal function in anesthetized dogs were observed.
In this study adult mongreal dogs of either sex weighing 11 to 13 Kg. were anesthetized with pentobarbital sodium, and 0.9 per cent saline solution containing p-aminohippuric acid and inulin were infused at a constant rate through the fermoral vein.
One hundred ten and 190 minutes after initiation of infusion, drugs to be studied were infused along with the saline solution for a forty minutes period.
Urine was collected through the polyethylene tubes inserted in both ureters. Systemic blood pressure was measured continuously from the femoral artery using a mercury manometer and kymograph. Urine volume, urinary sodium and potassium contents, renal plasma flow and glomerular filtration rate were calculated.
The dogs were divided into five groups according to drugs used; control(saline only), histamine 1.5μg group (histamine 1.59㎍/kg/min.), histamine 1.5㎍+chlorpheniramine group(histamine 1.5㎍/kg/min. with chlorpheniramine 0.2mg/kg. inital, 3㎍/kg/min.), histamine 5㎍ group(histamine 5㎍/kg/min.), histamine + chlopheniramine+furosemide group (histamine 5㎍/kg/min. with chlorpheniramine 0.2 mg/kg. initial, 3㎍/kg/min. and furosemide 33㎍/kg/min.). Each group consisted of five dogs.
The results were as follows.
1) In histamine 1.5㎍ group, the blood Pressure and urinary potassium, renal plasma flow (RPF) aud glomorular filtration rate (GFR) were slightly decreased whereas the urine volume and urinary sodium were rather significantly decreased.
In histamine 5㎍ group, the blood pressure declined significantly, and the urinary potassium, renal plasma flow (RPF), glomerular filtration rate (GFR) were more more significantly reduced than the former(histamine 1.5㎍ group), and the urine volume and urinary sodium were much more significantly reduced.
2) It appears that chlorpheniramine does not significantly influence the changes of renal function induced by histamine.
3. Furosemide increased the renal plasma flow (RPF), glomerular filtration rate(GFR), urinary sodium and potassium which were reduced by infusion of the histamine, but furosemide did not significantly influence the blood pressure, the decline of which was caused by the histamine infusion.
