Abstract

¸ñÀû: ³»Àμº Æ÷µµ¸·¿°ÀÇ ½ÇÇè ¸ðµ¨ÀÎ ½ÇÇèÀû ÀÚ°¡¸é¿ª Æ÷µµ¸·¿° ¸¶¿ì½º ¸ðµ¨ÀÇ ´«¿¡¼­ ¼¼Æ÷³» À§Çè½ÅÈ£¸¦ °¨ÁöÇÏ´Â ¿ªÇÒÀ» ÇÏ´Â Nod¾ç ¼ö¿ëü Àü»çüÀÇ ½Ã°£¿¡ µû¸¥ ¹ßÇöÀ» ¾Ë¾Æº¸°íÀÚ ÇÏ¿´´Ù.

°á°ú: ÀÚ°¡¸é¿ª À¯¹ß ÈÄ 2ÁÖ°¿¡ Nod¾ç ¼ö¿ëüÀÎ NLRP1, NLRP3, NOD1, NOD2ÀÇ Àü»çü ¹ßÇöÀÌ °¡Àå Áõ°¡ÇÏ¿´°í, ±× µÚ·Î Á¡ÁøÀûÀ¸ ·Î °¨¼ÒÇÏ¿´´Ù. ÀÌ Áß NLRP3ÀÇ ¹ßÇöÀÌ °¡Àå ³ô¾Ò´Ù. ¿°Áõ¼º »çÀÌÅäÄ«ÀÎÀÎ Interleukin-1¥âÀÇ ¹ßÇöµµ Æ÷µµ¸·¿° À¯¹ß ÈÄ 2ÁÖ°¿¡ °¡Àå ³ô¾Ò´Ù. ¸Á¸·ÀÇ ±¸Á¶Àû ÆÄ±«´Â Æ÷µµ¸·¿° À¯¹ß ÈÄ 3ÁÖ°¿¡ °¡Àå ½ÉÇÏ°Ô °üÂûµÇ¾ú´Ù.

°á·Ð: Nod¾ç ¼ö¿ëü Áß NLRP3°¡ ½ÇÇèÀû ÀÚ°¡¸é¿ª Æ÷µµ¸·¿°¿¡¼­ À§Çè½ÅÈ£¸¦ °¨ÁöÇÏ¿© ¿°Áõ ¹ÝÀÀÀ» ½ÃÀÛÇÏ´Â µ¥ Áß¿äÇÑ ¿ªÇÒÀ» ÇÒ °¡´É¼ºÀÌ ÀÖ´Ù.

Purpose: To evaluate the transcription pattern of Nod-like receptors (NLRs), the intracellular sensors, to detect danger signals in murine eyes with experimental autoimmune uveitis (EAU).

Methods: EAU was induced in B6 (C57BL/6) mice by subcutaneous injection of human interphotoreceptor retinoid binding protein and intraperitoneal injection of pertussis toxin. At 1, 2, and 3 weeks post-immunization, the eyeballs were extracted and subjected to histological and molecular assays using real-time reverse transcription polymerase chain reaction.

Results: The levels of nucleotide-binding oligomerization domain, Leucine rich Repeat and Pyrin domain 1 (NLRP1), NLRP3, nucleotide- binding oligomerization domain-containing protein 1 (NOD1), and NOD2 transcripts were increased at 2 weeks and gradually reduced thereafter. Notably, NLRP3 showed the highest expression in the eyes with EAU. Similarly, the transcript level of pro-inflammatory cytokine, interleukin-1¥â, increased and reached a peak at 2 weeks post-immunization. The retinal structure was severely damaged by inflammation at 3 weeks post-immunization.

Conclusions: Among NLRs, NLRP3 may induce inflammation in eyes after EAU immunization.
