Prapagation and Attenuation of Japaness Encephalitis Virus in Tissue Culture

Abstract


Japaness encephalitis virus (Nakayama strain) was cultrued through serial passages in HeLa, ammnion and chick-embryo fibroblast cells and its propagation process and induction of attenuation was assayed by the infectivity in mice.
1. It has been found tha tJapaness encephalitis virus was inactivated rapidly in cell-free medium incubated at 36℃ and completely disappeared at 43 hours of incubation.
2. The growth curve of Japanese encephalitis virus in Hela cells showed 24hours of latent period and reached peak at 4 days and continued to grow up to 12 days.
3. It was evident that Hela cells, amnion cells and chick-embryofibroblasts support the growth of Japnaese encephalitis virus. Teh cytopathic effect have been observed in the chick-embryo cells and Hela cells for first three or four serial passages. Teh infectivity to mice was reduced as the number of serial passage increases.
4. During the serial passage of the virus in HeLa cells, it has not been possible to prove pathogenicity in three-week old mice after seven serial passages, but seems to be attenuated, However, pathogenicity existed in one week old mice, This is interpreted to mean that the sucking mice are more sensitive to the virus than is the adults mice.
5. The kind of animal sera in the mdia used for virus culture did not affect on the cytopathic effect of the virus.
