Urinary Excretion of Pyridinoline Crosslink in Postmenopausal Women and Women with Premature Ovarian Failure

Abstract


Because the primary event following estrogen deficiency is an increase in bone resorption, a sensitive marker of bone resorption would be valuable to analyze the effect of menopause on bone turnover. The previous biochemical markers are
nonspecific,
insensitive and too cumbersome. The urinary excretion of pyridinoline(PYD) crosslink (PYD) crosslink has been suggested to be a sensitive and specific marker of bone resorption. The urinary PYD crosslinks were analyzed by CrosslinksTM immunoassay
in 30
postmenopausal women, 10 women with premature ovarian failure(POF) and 20 premenopausal women with normal menstrual cycle. Other biochemical parameters were also measured including urinary calcium/creatinine ratio, serum calcium, phosphorus,
alkaline
phosphatase, and osteocalcin. The mean urinary PYD crosslink and serum osteocalcin levels were significantly(p<0.05) increased in postmenopausal women and women with POF than in premenopausal women respectively. There were no significant
differences in
the mean serum alkaline phosphatase, calcium and phosphorus levels among 3 groups. The mean urinary calcium/creatinine ratio in postmenopausal women was significantly(p<0.05) higher than that of premenopausal women but no significant difference
in
this
ratio between premenopausal women and women with POF was noted. There were no significant correlations between urinary PYD crosslink and other biochemical parameters. Our data show that urinary PYD crosslink may be a promising marker of bone
turnover
which is accurate, inexpensive and convenient in the postmenopausal women and women with POF.
