Effect of Gonadotropins on the Proliferation of Human Endometrial Stromal Cells in Vitro

Abstract


It has been known that endometrial regeneration and proliferation is regulated by sex steroids, cytokines and various growth factors. The mechanisms responsible for such organized growth are not well understood yet. Recently, hCG/LH receptors
were
localized in endometrium by immunocytochemistry.
This study attempts to define the effect of human gonadotropins on the proliferation of endometrial stromal cells in vitro. Human endometrial stromal cells were prepared and cultured in serum-containing media. The effects of adding 7.5, 15, 30,
150mIU/ml of human menopausal gonadotropin(hMG) and follicle stimulating hormone(FSH), and 10, 100, 1,000, 10,000mIU/ml of human chorionic gonadotropin(hCG) on [3H]-thymidine incorporation were assessed.
Results indicated that FSH and hMG induced significant inhibition in stromal cell [3H]-thymidine uptake, resulting in 0.466 to 0.646-fold and 0.494 to 0.763-fold growth of control, respectively(p<0.05). Human chorionic gonadotropin did not
suppressed
stromal cell proliferation until the concentration of 1,000, 10,000mIU/ml were added (p<0.05) and was even stimulatory at 10mIU/ml although statistical significance was not found.
In conclusion, these data imply that human gonadotropins may inhibit human endometrial stromal cell proliferation though the dose-responsiveness did not achieve statistical significance, and this provides clues for understanding of direct actions
of
gonadotropins on the endometrial cells.
