Cardioprotective Effects of Estrogen and Enalapril on Monocrotaline-Induced Right Ventricular Hypertrophy in Ovariectomized Rats

Abstract


We previously demonstrated that ovariectomy (Ovx) accelerated the development of cardiac right ventricular (RV) hypertrophy in monocrotaline (MCT)-induced pulmonary hypertensive rats. Hypertrophied RV of the Ovx-MCT rats exhibited markedly higher mRNA levels of renin and angiotensinogen than that of the sham-operated female rats. Thus, in the present experiment, we investigated whether treatment of estrogen or enelapril, an angiotensin converting enzyme inhibitor, attenuates the progression of the RV hypertrophy. Female Sprague-Dawley rats (13 wk old) were subjected to bilateral Ovx or sham surgery. Two wk after the operation, a single subcutaneous injection of 60 mg MCT/kg or vehicle was given to Ovx and sham-operated female rats, and then 17-estradiol (50 g, sc., twice/wk) or enalapril (250 mg/L drinking water) was treated in Ovx rats for 4 wks. All sham-MCT and Ovx-MCT rats treated or untreated with estrogen and enalapril showed increased lung weights and morphological changes of severe pulmonary hypertension such as hypertrophied walls of small arteries, as well as atelectasis, interstitial thickening and inflammatory cell infiltration. In contrast, a marked RV hypertrophy was developed only in Ovx-MCT rats, but not in sham and estrogen or enalapril treated Ovx-MCT rats. Severe ultrastructural changes were shown in hypertrophied RV of Ovx-MCT rats such as disruption of intercalated disc, formation of hypercontraction band and myofibrilar fraying, but not in that of estrogen and enalapril treated Ovx-MCT rats. These results suggest that both estrogen and inhibition of the rennin-angiotensin system have a protective function against the development of cardiac remodeling in this rat model.
