Abstract
±èÄ¡·ÎºÎÅÍ °·ÂÇÑ H. pylori »ýÀ° ÀúÇØÈ°¼ºÀ» º¸ÀÌ´Â ±ÕÁÖ¸¦ ºÐ¸®, µ¿Á¤ÇÏ¿© Lb. plantarum NO1À¸·Î ¸í¸íÇÏ¿´´Ù. Lb. plantarum NO1Àº H. pylori»Ó¸¸ ¾Æ´Ï¶ó ±×¶÷ ¾ç¼º±Õ ¹× ±×¶÷ À½¼º±Õµé¿¡ ³ÐÀº ¹üÀ§ÀÇ ÀúÇØÈ°¼ºÀ» ³ªÅ¸³»¾ú´Ù. Lb. plantarum NO1ÀÇ ¹è¾ç »ó¡¾×À» H. pylori ¹è¾ç¾×¿¡ ÷°¡ÇÑ ÈÄ H. pyloriÀÇ urease Ȱ¼ºÀ» ÃøÁ¤ÇÑ °á°ú Lb. plantarum NO1ÀÇ °ÇÑ urease ¾ïÁ¦È°¼º(40~60% ÀúÇÏ)À» È®ÀÎÇÒ ¼ö ÀÖ¾ú´Ù. AGSÀ§¾Ï ¼¼Æ÷ÁÖ¿¡ H. pylori¸¦ ºÎÂø½ÃŲ ÈÄ Lb. plantarum NO1ÀÇ ¹è¾ç¾×À» ÷°¡ÇÏ¿© AGS¼¼Æ÷¿¡¼ H. pyloriÅ»Âø´ÉÀ» ÃøÁ¤ÇÑ °á°ú Lb. plantarum NO1Àº À¯»ê±Õ ¹è¾ç¾× ¹«Ã·°¡º¸´Ù 33% ÀÌ»ó ³ôÀº H. pylori Å»Âø´ÉÀ» ³ªÅ¸³»¾úÀ¸¸ç, ºñ±³±¸·Î »ç¿ëµÈ Lb. rhamnosus GG, Lb. sakei SI3¿¡ ºñÇØ ´õ ¿ì¼öÇÑ H. pyloriÅ»Âø´ÉÀ» ³ªÅ¸³»¾ú´Ù. ºÐ¸®±ÕÁÖÀÇ Àå³» »ýÁ¸¼º ¿©ºÎ È®ÀÎÀ» À§ÇÏ¿© ³»»ê¼º, ÀΰøÀ§¾×¿¡¼ 2½Ã°£µ¿¾È ó¸®ÇÑ °á°ú Lb. plantarum NO1ÀÌ Ãʱâ±Õ¼ö(109 CFU/ml)¸¦ À¯ÁöÇÏ¸é¼ ³ôÀº ÀúÇ×¼ºÀ» ³ªÅ¸³»¾ú´Ù. Oxgall ³óµµ 0.3%¿Í 0.5%ÀÇ Àΰø´ãÁó¿¡¼ 24½Ã°£ ó¸®ÇÑ ÈÄ¿¡µµ Ãʱâ±Õ¼ö(109 CFU/ml)¸¦ À¯ÁöÇÏ¿´´Ù. »Ó¸¸ ¾Æ´Ï¶ó ÀΰøÀ§¾×¿¡¼ »ýÁ¸ÇÑ ±ÕÁÖ¸¦ ¿¬¼ÓÀûÀ¸·Î Àΰø´ãÁóÀ¸·Î ó¸®ÇÏ¿´À» ¶§¿¡µµ ³ôÀº »ýÁ¸À²(108~109 CFU/ml)¸¦ À¯ÁöÇÏ¿´´Ù. Lb. plantarum NO1ÀÇ ¿ëÇ÷¼º ¹ÝÀÀ À¯¹« °á°ú ¿ëÇ÷¹ÝÀÀÀÌ ÀϾÁö ¾Ê¾ÒÀ¸¹Ç·Î ÀÎü¿¡ ¾ÈÀüÇÏ´Ù´Â °ÍÀ» °£Á¢ÀûÀ¸·Î È®ÀÎÇÒ ¼ö ÀÖ¾ú´Ù. º» ¿¬±¸¿¡¼ ±èÄ¡·ÎºÎÅÍ ºÐ¸®ÇÑ H. pylori¾ïÁ¦ À¯»ê±Õ Lb. plantarum NO1Àº Àå³»¿¡ »ýÁ¸ °¡´É¼ºµµ ³ôÀ¸¸ç, µ¿½Ã¿¡ À§¿¡¼ È¿°úÀûÀ¸·Î H. pylori¸¦ ¾ïÁ¦ ÇÒ ¼ö ÀÖÀ» °ÍÀ¸·Î ±â´ëµÇ¾îÁø´Ù.
One bacterium, which showed strong antagonistic activity against H. pylori KCCM 41756, was isolated from kimchi. The strain NO1 was designated as Lactobacillus plantarum NO1 based on Gram staining, biochemical properties, and 16S rRNA gene sequencing. The culture medium (2~4 ug/ml) of Lb. plantarum NO1 reduced (40~60%) the urease activity of H. pylori KCCM 41756. Lb. plantarum NO1 inhibited the binding of H. pylori to human gastric cancer cell line, AGS cells, by more than 33%. Lb. plantarum NO1 exhibited high viability (maintained initial viable cell count of 109 CFU/ml) in 0.05 M sodium phosphate buffer (pH 3.0) for 2 h, in artificial gastric juice for 2 h and in 0.3%, 0.5% oxgall for 24 h. Hemolysis phenomena did not observed when Lb. plantarum NO1 was incubated in the blood agar media. We concluded that Lb. plantarum NO1 can be a good candidate as a probiotic, harboring anti-H. pylori activity.
