Abstract


The indirect fluorescent antibody (IFA) test in the present study was developed by using delipidized adult Clonorchis sinensis particles as antigen fixed on microscope slide. The technic for preparation of antigen was based on the method for serodiagnosis of schistosomiasis (Camargo et al., 1966) with minor modifications.
Human sera from 38 cases of proven clonorchiasis, 91 cases of other parasitic diseases, 21 cases of parasites-negative liver diseases and 9 healthy controls were tested for the evaluation of this method.
In the results of the IFA test with the sera from clonorchiasis patients, the IFA titers obtained ranged between 1 : 8~1 : 256 with a distinct mode at 1 : 32. In the sera from other parasitic diseases, three samples from intradermal test positives to Paragonimus westermani antigen gave positive reactions titering 1 at 1 : 16 and 2 at 1 : 8, and one amebic liver abscess was positive at 1 : 8. All sera from parasites-negative liver diseases and heal-thy controls revealed negative for the test.
By the absorption of sera with delipidized adult Clonorchis sinensis particles, the sera from clonorchiasis converted to negative reaction and some sera, which had shown cross-reaction against Paragonimus antigen before absorption, became also negative against both antigens. In the sera from intradermal test positives to Paragonimus westermani antigen, the positive titer of 1 : 8 against Paragonimus antigen was absorbed and gave negative reaction, whereas titers of 1 : 16 or more remained intact when absorbed with C. sinensis particles. Some sera, which had shown cross-reaction against Clonorchis antigen, be-came negative with no change in titers for Paragonimus antigen.
The results indicate that IFA titers of 1 : 16 or more might reasonably be regarded as diagnostic of clonorchiasis, a titer of 1 : 8 as doubtful positive, and titers less than 1 : 8 as absence of this disease or nonspecific reaction.
