Effect of 2-Ethy-n-Caproic Acid on Cholesterol Induced Atherosclerosis in Rabbits

Abstract


The study was undertaken to observe the effect of 2-ethyl-n-caproic acid on development of experimental atherosclerosis in rabbits. The rabbits were placed on one of four diets containing combinations of lard, 2-ethyl-n-caproic acid and cholesterol for 10 weeks during which time serum alpha- and beta- lipoprotein cholesterol, free and _________ and phospholipids were determined at 2 week intervals. After termination of experiment, thoracic aorta of all animals was critically graded for the incidence and severity of atheroma and samples of aorta and liver were extracted for separation and analysis of tissue lipid fractions.
The result were summerized as follows:
1. The cholesterol ester, free cholesterol, and phospholipids and serum beta- lipoprotein and alpha- lipoprotein of rabbits fed with lard or lard and 2-ethyl-n-caproic acid showed no significant difference as compared with the values determined before the experimental diet started. Throughtout the whole experimental period, triglyceride content in beta- lipoprotein of rabbits fed with 2-ethyl-n-caproic acid showed slightly lower value than that of control animals.
2. During the period of cholesterol feeding, the serum lipoprotein lipid content showed a linear increase mainly due to elevation of beta- lipoprotein lipids.
3. In spite of the higher concentration of beta- lipoprotein cholesterol ester and free cholesterol in group fed with cholesterol and 2-ethyl-n-caproic acid than that fed with cholesterol, the development of atheromatosis in thoracic aorta was found to be far less in the former group.
4. The lipid content in liver and aorta revealed no noticeable difference between the control and 2-ethyl-n-caproic acid fed groups. In both group 2-ethyl-n-caproic acid fed groups. In both group fed with cholesterol and group with cholesterol and 2-ethyl-n-caproic acid, cholesterol ester free cholesterol and triglycerid were increaced than in control; especially in cholesterol ester.
