Contradicting results have been reported concerning the effect of diethyl ether on the activities of the pulmonary surfactant when diethyl ether was used as the general inhalation anesthetics to the rabbit, in vivo.
In the present study, an effort was made to elucidate a possible inhibitory effect of diethyl ether on the pulmonary surfactant, in vitro, and diethyl ether was added to the lung washings, obtained from the rabbit lung by washing out gently front the left lung twice with saline. in the ratios of 20:1, 12:1 and 6:1 (ratio of the lung washings vs diethyl ether).
The activities of the pulmonary surfactant were measured with the modified Langmuir-Wilhelmy balance on the 1st., 2nd., 3rd., 5th. and 7th. day after the addition of diethyl ether to the lung washings.
A control study was made in which the lung washings were stored at 4¡É for 7 days, and the activities of the pulmonary surfactant were measured at the same interval as in the experimental groups.
A comparison was made with the normal values of the minimal and maximal surface tensions of the surfactant as well as the stability index, and the results are summerized as follows :
1. The maximal and minimal surface tensions of the lung washings of the rabbit were 56.2 and 5.5 dyne/cm respectively, and the stability index was 1.64.
2. When diethyl ether was added to the lung washings in the ratio of 20:1 (lung washings vs diethyl ether), the activities of the pulmonary surfactant did not change from the control noticeably.
3. When the ratio of lung washings vs diethyl ether was made 12:1, a markedly decreased a activities of the pulmonary surfactant were observed throughout the entire experiment.
4. When the ratio of lung washings vs diethyl ether was further increased to 6:1, the activities of the pulmonary surfactant were decreased from the control markedly, but the decrease was similar to when in ratio was 12:1.
5. From the above results, it is concluded that diethyl ether produces a definite inhibitory effects on the activities of the pulmonary surfactant when used in a high concentration in vitro.
