Electrophoretic Patterns of Serum Alkaline Phosphatase Isoenzyme

Abstract


A very rapid method of cellulose acetate electrophoresis using New Dry Titan III YW Plate with a tiuperir~r visualization technique employing simultaneous coupling of a-naphthol AS-1~IX with a suitable diazo reagent (Fast blue RR), was performed for the identification of the tissue origin of scrum alkaline phosphatase in mainly liver disease, bone disease and normal personel. Combined with heat inactivation at 55´C for 30 minutes, specific for the placental enzyme, the hetercgenecity of serum alkaline phosphatase has been demonstrated. Normal adult serum contains predominantly liver type phosphatase (a~) with a small number of bone erzyme(a_/,3). In chilldhood, infancy anal prematxrre there appears that a bone enzyme is predominant in addition to the liver fraction. In late pregnancy, the placental enzyme (heat stable, a~/,3) is visualized predominantly.
From a study of seventeen cases of viral hepatitis and eight cases of obstructive jaundice, it is found that an ultraectopic fraction corresponding to the mobility of albumin is obtained in all cases, wtrilc thirty six cases of primary hepatoma and seven cases of liver abscess reveal ectopic (ar) fracti~,n. These t~vo .fractions are not well demonstrated in other publications but the author estim~rte´ the origin of these two kinds of ectopic fractions to be the liver in some particular diseases whatever the mechanism of production of the particular isoenzyme involve.
The bone disease reveals primarily the bone fraction. It is also interesting to see the ectopic fractir>n in the liver tissue of five months gestation arld a-fetoprotein antiserum. There is no change irr the placental and ultraectopic fraction by heat (35C, 30 minutes). The serum may be stored at 4-6C for seven days without remarkable changes in the patterns of alkaline phosphastase isoenzyme.
The final fractions can be scanned and quantitated by densitometer.
