Abstract
Niclofolan (5, 5¢¥-dichloro-2, 2¢¥-dihydroxy-3, 3¢¥-dinitro biphenyl) decreases tonus of isolated intestine, but mechanism of decreased tonus induced by niclofolan has not been proved.
To observe the machanism, the effect of acetylcholine (2¡¿10^(-6)g/ml), calcium chloride (2¡¿10^(-4)g/ml) and barium chloride (4¡¿10^(-4)g/ml) were otserved on the isolated rat duodenum suspended to Tyrode¢¥s solution at 38¡É.
Some investigators, reported that acetylcholine and barium chloride show phasic contraction (PC) due to Ca-release and tonic contraction(TC) maintained by energy-dependent Ca-influx, particularly acetylcholine utilizes easily releasable Ca, and barium chloride utilizes easily releasable ca and less easily releasable Ca.
To observe mechanism of niclofolan-induced action particularly in relation to calcium, this suggestion was applied.
Acetylcholine and barium chloride-induced PC and TC followed 30 minute later by niclofolan were depressed.
Acetylcholine-induced TC followed 20 minute later by niclofolan or by calcium-free without niclofolan was more markedly depressed than PC.
But barium chloride-induced PC and TC followed 20 minute later by niclofolan were depressed and by calcium-free without niclofolan were not depressed.
In the calcium-free media barium chloride-induced TC followed 10 minute by niclofolan was more significantly depressed than PC.
Addition of calcium chloride made to restore from decrease in tonus induced by calcium-free, but the restoratioa due to calcium chloride was sinificantly inhibited by the presence of niclofolan.
These results suggest that utilization of releasable calium might be inhibited by niclofolan, but these would not prove either inhibition of release from calcium pool or direct inhibition to contractile protein.
