Studies of Ceruloplasmin Activity in the Serum of the Patient with Pulmonary Tuberculosis in Korea

Abstract


Ceruloplasmin, the blue copper-containing protein of serum, was first isolated in 1948 by Holmberg and Laurell. They characterized this protein as an alpha 2-globulin with a copper content of approximately 0. 32 percent, a molecular weight of about 151, 000 and 8 atoms of copper molecules.
Holmberg and Laurell demonstrated that such patients most of the copper is associated with the globulin fraction and that there is a high degree of correlation between the copper content of the sera and the oxidase activity. In a Pregnant woman, the hypercupremia is due mainly to an increase in ceruloplasmin, although there is a modest increase in the direct reacting fraction as well.
It is now known that hypocupremia is present in most patients with the nephrotic syndrome.
In Wilson´s disease it has been demonstrated that there is a marked decrease in the indirect reacting fraction of plasma copper and an increase in the direct reacting fraction.
The purpose of this paper is to present the results of measurements of ceruloplasmin activity in the serum of the normal human, the patient with pulmonary tuberculosis and the normal rabbit.
I have also measured the ceruloplasmin concentration in the serum of patient with pulmonary tuberculosis and normal rabbits after the administration of ascorbic acid, pyridoxine and thiamine.
The ceruloplasmin activity was determined according to the method of Henry, Chiamori and Segalove. The result is shown as follows.
1. The ceruloplasmin activity in the serum of the Korean adult is as follows: male, 282 units; female, 329 units.
2. The ceruloplasmin activity in the serum of patients with pulmonary tuberculosis is different in each one accordig to the severity of diseases.
a. The minimal pulmonary tuberculosis: male, 321 units female, 355 units.
b. The mcderately advanced pulmonary. tuberculosis: male, 400 units; female, 483 units.
c. The far advanced pulmonary tuberculosis: male, 505 units; female, 633 units.
The ceruloplasmin activity is the serum of patients with pulmonary tuberculosis above listed in a, b and c was found increasing according to the severity of diseases.
3. The ceruloplasmin activity in the serum of patients with pulmonary tuberculosis was markedly depressed by the administration of pyridoxine, ascorbic acid and thiamine. The supression of ceruloplasmin activity is more marked by the administration of pyridoxine than ascorbic acid and thiamine.
4. The activity of ceruloplasmin in the rabbits was markedly depressed according to the changes of time and days of the administration of pyridoxine, thiamine and ascorbic acid. The depression of ceruloplasmin activity is more marked by the administration of pyridoxine than ascorbic acid and thiamine.
