Abstract


Like all other antimicrobial agents, streptomycin produces a variety of undesirable side effects. Some
of these are due to hypersensitization; others derive from the direct toxic effect of the antibiotic; still others result from biological complications in the host and are not associated with the toxicity of the drug or hypersensitivity to it.
For the purpose of investigating the antigenicity of streptomycin, streptomycin was emulsified in complete Freund´s adjuvant and used for sensitization of rabbits and guinea pigs by injecting intradermally and intramuscularly. All experimental animals were treated again with the same sensitizing agent by smearing on the scarified dorsal shin trice a week for 4 weeks. After a 21 day rest period, the animals were bled partially, and the sera were tested serologically; precipitation, complement fixation, complement consumption, indirect hemagglutination, hemagglutination inhibition, modified hemolytic reactions, and Prausnitz-Kuestner reactions.
Four days after the partial bleeding, the same animals were also tested for hypersensitivity to streptomycin by intradermal skin tests and anaphylactic shock with a solution of the sensitizer. The results obtained were as follows:
1. The sera of sensitized animals showed negative precipitation and complement fixation reactions, but showed positive complement consumption test and the other serological procedures tested.
2. The positive reactions of the blocking test by hemagglutination inhibition test may indicate the production of the specific circulating antibodies to streptomycin in the animals.
3. PrausniL-Kuestner reaction revealed negative results but showed positive results by direct intradermal skin tests to streptomycin in the animals.
