Abstract
Both Mg¢¥ and Na+-K+ ATPases activities in Eomogenate obtained from renal cortex of rat was measured in vitro under the variety of conditions. The effect of ethacrynic acid on the activities of both ATPases was investigated and the results were summarized as follows.
1. The Na+-K+ ATPase activity was depressed in a greater extent than the Mg¢¥ ATPase by ethacrynic acid, especially in the presence of lower K+ concentration. And then mercury chloride depressed both Mg" and Na+-K+ ATPases activities.
2. The inhibition of both Mg" and Na+-K+ ATPases activities by ethacrynic acid or mercury chloride was prevented by an addition of cysteine in the medium.
3. Reactivation of Na+-K+ ATPase by cysteine which had been depressed by ethacrynic acid was markedly reduced in the absence of K+.
4. The inhibition of Na+-K+ ATPase by ethacrynic acid was more pronounced by the longer preincubation time while reversal of the inhibition by cysteine was not complete.
