Abstract


In order to study histochemically the changes in glycogen distribution and phosphorylase activity in the muscle fibers of immobilized skeletal muscles, one of the hind limb of rat was fixed with plaster cast and observed the glycogen distribution and phosphorylase activity in the muscle fibers of gastrocnemius, semitendinosus,. semimembranosus and soleus at one, two, four, six, eight, ten and twelve weeks after fixation and at three and four weeks aftre release of cast, and also observed those in the muscle fibers of the non fixed limb.
The results obtained were as follows:
1. The glycogen distribution and phosphorylase activity in the muscle fibers of gastrocnemius, semi­tendinosus and semimembranosus of normal rats were different in each types of fibers, i.e., more glycogen in medium and large fibers and less in small fibers, and high phosphorylase activity in large and medium fibers and low in small fibers. The glycogen distribution in the muscle fibers of soleus was in moderate amount and phosphorylase activity was relatively low.
2. The glycogen distribution in large and medium fibers of gastrocnemius, semitendinosus and semi­membranosus, and muscle fibers of soleus was gradually decreased from one to twelve weeks after fixation but the glycogen distribution in small fibers of those muscles was slightly increased from one to eight weeks and again decreased in ten and twelve weeks after fixation. The glycogen content in the muscles which had been reduced during immobilization showed increase again after the release of cast.
3. The phosphorylase activity in the muscle fibers of skeletal muscle in immobilized limb showed gradual decrease from one to ten weeks and almost disappeared in twelve weeks after fixation but increased again after release of cast.
4. The glycogen distribution and phosphorylase activity in the muscle fibers of skeletal muscles in non fixed limb of experimental rat were different in each muscles and each types of fibers but the glycogen distribution and phosphorylase activity showed a tendency to gradual decrease in the course of experiment.
