Abstract


In order to investigate histochemical properties and sequential changes of the mucosubstances in the mucosa of the proventriculus and gizzard, observations were made on chick embryo at 7,10, 13,16 and 19 days of incubation and also chicken at 1, 4, 7 and 10 days after hatching. The mucosubstances were stained with alcian blue (pH 1.0, 1.5), PAS, alcian blue (pH 1.0, 2.5) - PAS and aldehyde fuchsin (pH 1.7)¡©alcian blue (pH 2.5) stainings.
The results obtained were as follows:
1. The mucosubstances in the surface epithelial cells and glandular cells of proventriculus in developing chick were composed of acid and neutral mucin and were more acidic in 7-13 day chick embryos, but more neutral from 16 and 19 day chick embryos to 10 day after hatching. The mucosubstances in the pit epithelial cells of proventriculus contained only a small amount of neutral mucin before hat¡©ching but were composed of mainly neutral and some acidic mucin after hatching. Generally, the content of neutral mucin in the proventricular mucosa of developing chick gradually became increased after hatching.
2. The acid mucosubstances in proventriculus of developing chick were mainly sulfated mucin, and also appeared to contain nonsulfated mucin in the chick after hatching.
3.The mucosubstances in the surface epithelial cells and glandular cells of the mucosa of gizzard in developing chick contained acid and neutral mucin, being predominantly acidic in surface epithelial cells, while neutral mucin was predominants in glandular cells.
