Abstract


The author investigated the control state of the counting process of blood leukocytes in our laboratory, and found that there were a few sources of error which seriously affected the counting process. In this study the author tried to evaluate the effectiveness of the experimental designs applied by him to the investigation, and found that they were quite satisfactory in such observation as this.
Summary
1. There was no difference between the two chambers and pipettes equipped in our laboratory, and found them still useful. Batter results were obtained when the chamber and pipette belonging to the same set were used.
2. It was more safe to use the 4th and 5th drops after discarding 3 and 5 drops of diluted blood from the pipette, respectively.
3. In the counting procedure the two types of dilutions of blood, that is, 20:1 and 10:1, made no difference to the results.
4. It was found that the most serious error source was in the blood samples collected in the containers, and that it was utmost important to mix the entire blood sample to keep equal distribution of the leukocytes suspended in the blood.
5. The two technicians working at the hematology section at present seemed to be well qualified.
6. In this study some variations and errors concerning leukocyte counts were estimated.
1) The S. D. (standard deviation) of repaated measurements of the same blood sample was estimated at about 400/cu mm.
2) The S. D. of variation in leukocyte counts between blood samples obtained from an individual at the same time was estimated at about 700/cu mm and the S. D. of their leukocyte counts at about 800/cu mm.
3) The S. D. of variation in leukocyte counts between healthy adults was estimated at about 1100/cu mm and the S. D. of their leukocyte counts at about 1400/cu mm.
4) The mean and the 95% range of leukocyte counts in healthy adults were estimated at 7000/cu mm and 4500-10000/cu mm, respectively.
