Abstract


The distribution of eosinophilic leucocytes was studied in the peripheral blood, intraperitoneal fluid and various kinds of organs and tissues, after eosinopenia was experiemtanlly induced by several methods. Also, in the state of peripheral eosinopenia, variations of the chemical components in the eosinophilic granules and cytoplasm of the cells were observed from the viewpoint of histochemical methods.
The maximal decrease of 50-851 of eosinophils appeared in 4-6 hours after an injection of ACTH (5, 10, 20 u/kg), cortisone-acetate (1, 5, 10 mg/kg) and adrenalin (0.1, 0.5, 1.0 mg/kg) into the peritoneal cavity of guinea pigs injected separately.
After total adrenalectomy, injection with either cortisone-acetate or adrenalin, showed that the eosinophils decreased in contrast to that of the non-adrenalectomized animals in which injection of cortisone-acetate and adrenalin at the same time had no effect in decreasing the eosinophil count.
In a group with-cervical transection of the spinal cord, injection of cortisone-acetate or adrenalin made no difference in the decreasing rate of eosinophils with that of non-transected group.
While cortisone-acetate made a 68% drop in eosinophils 6 hours after an injection adrenalin made a-31% drop and both cortisone-acetate and adrenalin injected together made a 64% drop in eosinophils in the animals with reticulo-endothelial system (RES) blocking. The degree of drop in the animals with non-RES blocking is greater than that of RES-blocked animals.
Stress caused the most severe drop of eosinophils, as a 100% drop occurred in 8 hours after stress was given, and compulsive stress showed a more marked drop than the stress of insomnia.
In all the above experimental data, the drop and rise of eosinophils in the periheral blood was earlier than that in the peritoneal fluid, but there was no difference in number of eosinophils.
No significant difference in the number of eosinophils was observed between the 5 hour incubated healthy adult blood mixed with ACTH (100 mg%), cortisone-acetate (100 mg%) and adrenalin (5mg%) and in the control group mixed with NaCl in vitro.
In the most severely induced experimental eosinopenia the distribution of eosinophils was observed most commonly as a massive clustering in the peribronchial and peribronchiolar connective tissue, the lumen of the bronchi, and next. to the bronchial epithelial cells. In the perivascular.tissue, septum of alveoli, lamina propria of small intestine and intestinal: epithelium, (especially Paneth cells), the eosinophils were also appeared in massive grouping.
Eosinophils in the bone marrow increased-´20-30% in the state of eosinopenia;-and a slight increase of eosinophils was also observed in the red pulp of the spleen. Interlobular fibrous tissue of, the liver and skin revealed extremely rare eosinophils, and there were no eosinophils detectable in the kidney, adrenal-gland and hypophysis.
In the various types of eosinopenia, the reaction of eosinophils to the several kinds of histochemical procedures was observed and the following results were obtained:
Periodic Acid Schiff reaction (PAS reaction) of the cytoplasm of the eosinophils revealed faint staining and a fine reticulum and the proper granules of the eosinophils were PAS negative as well as those in the control group.
The proper granules of the eosinophils appeared as positive in the staining of ribonucleic acid(RNA) which disappeared by the action of ribonuclease. Desoxyribonucleic acid (DNA) in the nucleus of eosinophils decreased with arise of RNA in the same nucleus.
Alkaline phosphatase reaction was strongly positive giving granules of chocolate color in both the proper granules and cytoplasm of the eosinophils.
The above described histochemical reaction was much stronger in the peritoneal fluid, the organs and tissues, than in the eosinophils of the peripheral blood, especially in the group where stress was given.
When the decreased eosinophils returned to a normal level, the histochemicai properties also returned to normal.
In the test tubes, (in vitro) the experimental group showed no difference in the histochemical characters of the control group mixed with NaCl.
