動物體內에서의 Klebsiella Pnenmoniae Type 2 Phage의 運命에 關한 硏究

Abstract

正常狀態에 있는 사람 또는 動物의 血淸 및 其他 體液이 試驗管內에서 Klebsiella Pneumoniae Type 2 phage에 미치는 影響을 檢査하여 다음과 같은 結果를 얻었다.
個差는 甚하였으나 사람과 動物의 血淸은 phage를 中和하였으며 一部 phage는 이에 抵抗하여 殘存하였다.
사람의 腹水에는 phage中和能力이 相當程度 있었으나 尿, 침, 髓液, 膿, ○○, 糞 그리고 動物의 尿, 침, 糞에는 없었다.
37℃에 12時間 放置한 馬血淸의 phage中和能力은 新鮮 馬血淸의 그것에 比해 큰 差異는 없었으나 時間이 지남에 따라 漸減되었으며 8日에 이르러서는 거의 消滅하였다.
사람의 腹水를 37℃에 放置한 結果도 馬血淸의 境遇와 大略 비슷하였으며 8∼9日에 phage 中和能力은 거의 消滅되었다.
소, 말, 개 等의 血淸 그리고 사람의 腹水를 20℃에 保存하면 1∼2日에는 phage 中和能力에 큰 變動은 없었으나 5∼7日에 約 50% 減少, 15∼16日에는 거의 消滅되었다.
소, 말, 개 等의 phage 中和能力은 50℃에 1時間 加熱로 減少되기 始作하여 56∼58℃ 1時間에 70∼80%, 그리고 72℃ 1時間에 完全 消滅하였다.
馬血淸의 phage 中和能力에 가장 適合한 pH는 7.0이었으며, 6.5∼7.5에서도 큰 變動은 없었다.
The author has studied the effects of sera and various body fluids of normal healthy persons and animals upon the activity of klebsiella pneumoniae type 2 phage in the test tube, and obtained the following results.
The serum of normal healthy persons and animals neutralized the activity of phage. However, the Phage neutralizing activity of serum showed marked individual and species difference and a part of phage remained a1ive and resistant to the action of sera.
The human ascitic fluid neutralized the phage, to a marked degree, but the other human body fluids, such as urine, saliva, spinal fluid, pus, bile and stool and animal·body fluids, e. g. urine, saliva and stool had no effect on the activity of the phage.
The phage neutralizing activity of horse serum treated at 37℃ for twelve hours did not show any significant difference from that of fresh horse serum. However, the neutralizing activity disappeared gradually as time elapsed and it was almost completely destroyed by treating the serum 37℃ for 8 days.
The phage neutralizing power of human ascitic fluid also disappeared gradually by keepiug at 37℃ and the response was similar to that of horse serum i.e. by keeping the ascites at 37℃ for 8~9 days, the neutralizing activity was almost nil.
On keeping the sera of the on, horse and dog· and human ascites at 20´C, the phag neutralizing activities of these sample showed no appreciable changes for 1~2 days. However,´by 5~7 days, the activity dropped to about 50% of the original ones and, by keeping for 15∼16 days the neutralizing activity was almost completely destroyed. The phage neutralizing powers of sera of the ox, horse and dog and human ascites were quite stable at 4℃ for 3∼5 days and at -4O℃ for about two months.
The phage neutralizing activity of horse serum began to decrease by heating at 50℃ for one hour and about 70~80% of the activity disappeared at 56℃~58℃ for one hour and, was completely destroyed at 72℃ for one hour.
The most suitable pH for the action of phage neutralization of horse serum was 7. 0 and, between pH 6.5∼7.5, no great change was found.
