Abstract

The mechanisms of the hypotension and bradycardia produced by 5-hydroxytryptamine ( 5-HT, 50㎍/kg, i.v.) in the rabbit were investigated.
1) The latent period from the time of injection until the onset of the bradycardia was different according to routes of administration. It was from 1.0 to 5.0 seconds by car vein injection. The onset of the bradycardia was immediate when given directly into the right atrium or the lift ventricle, but it was slower when injected into the carotid artery-than into the car vein. Increasing doses of 5-HT(200㎍/kg) injected into the carotid artery elicited miiediate bradycardia.
2) Phentlbiguanide (100㎍/kg) elicited transitory hypotension and bradycardia as 50HT did. The onset of the bradycardia was immeiated when given either into the lift ventrcle, the fight atrium or the carorid artery. It required from 0.5 to 3.4 seconds by the car vein injection.
3) Protoveratrine(1-2mg/kg) elicited hypotension and bradycardia, which persisted for a period of 20 to 30 minutes. In the presence of protoveratrine the hypotension by 5-HT was attenuated but the bradycardia was intensified.
4) Treatment of rabbits with lidocaine, a local anesthetic, abolished the bradycardia by 5-HT and attenuated the hypotension.
5) In chlorisondamine-treated rabbits 5-HT elicited pressor effect and the bradycardia by 5-HT was not apparent. The pressor effect was inhibited by methysergide.
6) 5-HT elicited no hypotensive effect in the majority of rabbits treated\ either of bretylium, guanethidine or phenoxybenzamine, but produced slight rise of blood pressure. The bradycardia by 5-HT was not affected either of these agents.
7) Reserpine-treatment of rabbits did not affect the bradycardia by 5-HT but the hypotension was attenuated or reversed to tile weak Pressor effect.
8) Rabbits treated with JB-516 and DOPA responded with the bradycaidia to 3-HT, but showed no hypotension.
9) From the above results it was assumed that (1) the hypotension and bradfcardia by 5-HT in the rabbit were reflexive in nature, (2) the sensory recep색s in the intrathoracic area participated in this reflex, (3) the hypotension was due to inhibition of the central sympathetic tone, (4) the bradycardia was doe to elevation of the central vagal tone. In other words,5-07 seemed to elicit "Bezold-Jarish Reflex" in rabbits.
