Some Autonomic Drugs on the Intraocular Pressure and on the Isolated Pupillary Muscles of the Rabbit

Abstract


Effects of some autonomic drugs on the intraocular pressure and on the isolated pupillary muscles of the rabbit were investigated.
1) A method for registration of the intraocular pressure was described. The pressure was 217.4± 6.5mmH₂O(mean of 40 cases± S.E.) by this method.
2) Norepisephrine, Isopfoterenol and 5-Hydroxytryptamine, when given intravenously produced. an immediate rise or fall of the intraocular pressure. corresponding to the changee in the arterial blood pressure.
3) Norepineplirine and angioiensin, when given into the lingual artery, produced a fail of the intraocular pressure ipsilateral to the side of injection, regardless of the changes in the arterial blood pressure.
4) DMPP and McN-A-343: whatever the route of administration was produced a rise of the intraocular pressure, being independent from a fall of the arterial blood pressure.
5) 5-HT produced a fall of the intraocular pressure, even when the ·fall of the blood pressure was prevented by phenotybenzamine or chlorisondamine.
6) The above results concerning the intraocular pressure indicated that·the changes in the intraocular pressure by the autonomic drugs and 5-HT were partly-Independent from the arterial blood pressure level. It was discussed that responses of the intrascleral blood vessel to drugs may play a role in eliciting changes in the intraocular pressure.
7) Methods for preparing isolated preparations of the sphincter muscle and the radial muscle of rabbits were presented.
8) Acetylcholine contracted the sphincter muscle but not the tradial muscle.
9) Sympnthomimetic amines (epinephrime, norepinephrime and isoprote-renol) relaxed the sphincter muscle being contracted by acetylcholine, which was prevented by propranolol, The radial muscle responded with contracture to epinephrine, norepinephrine. and prevented ·by phenonybenzamine. Smaller doses of isoproterenol produced a relaxation of the radial muscle which was prevented by propranolol.
10) DMPP and 5-HT Produced contraction of the both muscles. The contraction of the sphincter muscle was prevented by either of hexamethonium or atropine. The contraction by 5-HT was also prevented by methrsergide.
11) McN-A-343 relaxed the sphincter muscle being contracted by acetylcholine, but produced a slight contraction or no effect on the radial muscle.
12) The results concerning the pupillary muscles it dicated the presence of alpha and beta type of the adrenotropic receptor in the radial muscle and beta type in the sphincter muscle. It was suggested the presence of specific sites of action for autonomic ganglionic stimulants in the pupillary muscles of the rabbit.
