Abstract


The effect of INH on the biosynthesis of- hepatic NAD from nicotinamide have been investigated- in mouse by following- the changes in the level of NAD and. observing the incorporation of nicotinamide7-14C into. the precursors of NAD biosynthesis, namely, nicotinic acid mononucleotide, desamido-NAD and NAD.
1. INH(GOmg/kg), given intraperitoneally,,either 8 hours : previously orconcomitantly with the administration of nicotinamide, inhibited markedly the increase in.hepatictNAD resulting from the administration of nicotinamide( mg/kg) The INH itself, however, when given alone, did not-cause any change a the level of hepatic NAD.
2. the analysis for the incorporation of nicotinamide 7-14C by Dowex 1-formate:-column chromatography and by paper selectrophoresis showed , that the incorporation of nicotinamide into NAD increased with time from 2 to 8 hours after the administration of nicotinamide, while that into desamido-NAD reached maximum at 4 hours and decreased thereafter.
The concomitant administrationn of INH caused a marked decrease in the incorporation into NAD fraction of nicotinamide at anytime following nicotinamide treatment. However, the INH given concomitantly with nicotinamide brought about a increase in the incorporation into desamido-NAD fraction at a and 8 hours with a decrease at 4 hours after the administration of nicotinamide.
From these results, it might be concluded that INH inhibits a hepatic NAD biosynthesis from nicotinamide by blocking the conversion of desamido NAD to NAD in the final steps of NAD biosynthetic pathway.
