Abstract


1. When slices of skin from young rats were incubated at 37°C with Krebs-Ringer phosphate buffer medium containing nicotinic acid-7-14C and glucose, the radioactivity of nicotinic acid disappeared gradually from the medium and was found to be taken up by skin slices. The uptake of nicotinic acid by skin slices was markedly inhibited by sodium cyanide, p-chloromercuribenzoate and 2,4-dinitrophenol.
The uptake of nicotinamide by skin slices, on the other hand, was found to be much less than those of nicotinic acid.
2. In the presence of ATP and glutamine, most of the nicotinic acid taken up by skin slices was incorporated into NAD and the intermediates of NAD biosynthesis such as nicotinic acid mononucleotide, deamido-NAD, and nicotinic acid riboside, etc. Free nicotinic acid-14C, however, was barely detectable in the skin slices.
3. The process by which skin slices incorporate nicotinic acid into NAD and the biosynthetic intermediates was found to be strongly ATP dependent, and in the absence of ATP the incorporation of nicotinic acid into NAD and the intermediates was markedly lowered. The absence of glutamine in the -incubation medium also lowered the incorporation, but was not so pronounced as compared to the absence of ATP.
4. These results were interpreted as indicating that nicotinic, acid is actively taken up by rat skin slices and that a considerable portion of the nicotinic acid taken up is converted to NAD via a biosynthetic pathway which is operating in the other tissues like erythrocytes and liver.
