Abstract


1. Quinolinic acid was converted to niacin ribonucleotide with the release of CO2 by rabbit liver slices during incubation at 37°C in Krebs-Ringer phosphate medium. The process was markedly enhanced by the addition of glucose and ATP to the medium. In comparison, the incorporation of nicotinic acid to the tissue nucleotides was much greater than that of quinolinic acid, and was also enhanced by glucose and ATP in much greater magnitude (2 to 4-fold)than of quinolinic acid. On the other hand, the addition of ribose-5-phosphate in the presence of glucose and ATP brought about a lesser increase in the incorporation into tissue nucleotides of both compounds than when only glucose and ATP were added without ribose-5-phosphate.
2. The addition of each of nicotinic acid and nicotinamide (1.0 mM) to the medium containing glucose and ATP led to a little suppression (by 14%) on the incorporation of quinolinic acid into the tissue nucleotides.
3. From these results, it might be suggested that the cell permeability of quinolinic acid is far less than of nicotinic acid, and that the conversion to NAD of quinolinic acid from the extracellular ´source plays a minor physiological role as compared with the contribution of nicotinic acid in NAD biosynthesis.
