Abstract


1. With the view of improving the efficiency of ATP-regenerating system with 3-phosphoglycerate and rabbit muscle extract, a study was made of the enzyme composition of the muscle extract, and of the relative efficacy of adenine. nucleotides added to this system.
2. In addition to the glycolytic enzymes which catalyze the reaction sequence from 3-phosphoglycerate to lactate, adenylate kinase has been shown to be present in the muscle extract. The activities of these enzymes were found to be sufficient for the efficient regeneration of ATP.
3. The rate of oxidation of reduced NAD by the muscle extract in the presence of 3-phospboglycerate as a measure of the efficiency of ATP regeneration was more marked with the addition of AMP plus ATP or of AMP alone than with ADP or ATP added to the system.
4. A reduction in ATP regeneration was observed when an increased amount of ADP or ATP was added to the system and incubated for the period of more than ten minutes. With AMP, however, this phenomenon was not observed.
5. These results suggest that with this ATP-regenerating system care must be exercised that more than catalytic amounts of ATP should not be added, and that the addition of AMP plus ATP or of AMP alone might be much more efficient for the regeneration of ATP than with the addition of
