Abstract


1. The isozymes of glutamic-oxaloacetic transaminase from chicken´ liver supernatant and mitochondrial fraction were studied with respect to their activity changes during development at 5 to 10 days after hatching, and some of their enzymic properties were investigated in comparison with those of rat liver isozymes.
2. The activities off the supernatant and mitochondrial isozymes did not show any change during growth when expressed in the wet weight of tissue, and it was found that 95% of the activity exists in the supernatant fraction. However, when expressed in specific activity per mg protein, a gradual increase with the growth of chicken was noted in the activities of the supernatant and mitochondrial fractions as well as of the whole homogenate.In adult chicken the specific activities of whole, homogenate and supernatant fraction decreased yet with the continued increase in the .mitochondrial fraction with 15% of the total activities found in the mitochondrial fraction.
3. In marked contrast to the rat liver isozymes, chicken liver supernatant and mitochondrial isozymes showed the similar properties in DEAE-cellulose, column chromatography in the responses to temperature and pH changes, and in their apparent Michaelis constants for L-aspartate. However, both isozymes of chicken liver resembled the rat liver cationic mitochondrial isozyme in their pH dependence and chromatographicaI behavior on DEAEcellulose column, whereas they were similar to the rat liver anionic supernatant isozyme with respect to their stability toward temperature and in their apparent Michaelis constants for L-aspartate.
4. From these results it is suggested that chicken liver contains mainly one isozyme of glutamic oxaloacetic transaminase in contrast to two isozymes in rat liver.
