Abstract


A study was made off the incorporation of DL-serine-3-14C into globinr. and heme of chicken nucleated erythrocytes, in vitro, the results of which are presented below.
1. In the presence of glucose and 18 other L-amino acids intact wholered blood cells actively incorporated serine-14C into the globin. However, the hemolyzed red cells showed a markedly decreased incorporation. The incorporations into the heme was not influenced to any extent by hemolysis.
2. The cytoplasmic fraction obtained, by differential centrifugation to remove cell nuclei and cell membrane debris actively incorporated serine-14C into the globin to the same extent´ as by intact cells upon the supplementation of glutathione, 18 other amino acids and of an ATP-generating system. Therequirement for these supplements, especially for ATP generating system wasdemonstrated.
The synthesis of the heme by the cytoplasmic fraction was almost similar irrespective of the presence of the supplementation.
3. From these results it might be concluded that the biosynthesis of protein in chicken nucleated erythrocytes´ is mainly a function of ribosome ire the cytoplasm as demonstratedfor non-nucleated reticulocytes and very small portion of protein is synthesized in the cell nuclei.
