Abstract


Sympathomimetic effects of -bretylium were investigated in blood pressure response of rabbit and tortoise and in isolated atria of tortoise. 1) Bretylium licited no pressor response and no cardioacceleration in whole rabbit. In spinal rabbit, however, bretylium produced pressor response and cardioaccelerat on. bretylium produced pressor response in tortoise. 2) The pressor response to bretylium in spinal rabbit and: tortoise was more pronounced within 2 to 3 hours after intravenous injection of reserpine. 3) The pressor response to brety1iumn was not elicited when the animals were pretreated with reserpine. The reserpine-pretreated spinal rabbit, however,responded with pressure rise after norepinephrine infusion. 4) The isolated atria of the tortoise responded to bretylium with increase of contraction amplitude. This effect could not to observed in the presence of nethalide or cocaine. The atrial preparations made from the reserpine pretreated tortoise showed much weaker response to bretylium than control atria. When the preparations were incubated with norepinephrine, the contractile response to bretylium increased. 5) From these it is confirmed in rabbit and tortoise that bretylium exerts its sympathomimetic effects by releasing catecholamines.
