Abstract


Histological and histochemical studies were made on the kidney of the turtle(Amyda japonica) and followings were the summary of the results:
Gross examination of the turtle kidney showed furrows, external convolutions and internal lobations. Connective tissue of the capsule could be tracedinto the furrow as the septum between the adjacent convolutions, and it contained many blood vessels, some cellular elements, and abundant collagenous fiber bundles. Some smooth muscle cells were frequently found in the connective tissue near the median ventral area of the kidney with no significant relationship with surrounding structuress.
Renal corpuscles were arranged in one row laterall to the large vessels medially located. Each nephron, starting from. the. Bowman´s capsule as a short neck, included proximal tubule, short intermediate segment which was perhaps corresponded to Henle´s loop in mammals, and distal convoluted tubule near the renal corpuscle. Continuous to distal tubules were a branch of the collecting segment, which ran parallel with proximal tubules, and connected with some angle to collecting duct located almost midway between glomerulus and peripheral renal capsule.
The cells of the proximal tubule were characterized, by having acidophilic cytoplasm and prominent brush border and containing abundant mitochondria of granular and filairentous shape.
In the intermediate portion, small cuboidal cells, typically ciliated, badd basophilic cytoplasm and very few mitochondria.
Relatively long distal tubules were composed of cuboidal cells which had basophilic cytoplasm and mainly filamentous mitochondria arranged vertical to the basal cell surfaces showing basal stria.
In a collecting poriton were found ordinary epithelial cells and mitochondria rich-cells, and in the more distally located collecting duct mucous cells were also present.
In the same section, lobes with contracted tubules and vessels as well as one with dilated were sometimes found.
Saliva-resistant PAS positive materials in the lumina of tubules tended to increase from proximal tubule to collecting duct, in which these materials reached maximal concentration. These PAS-substances were identified as acid mucopolysaccharide by colloidal iron staining and metachromasia with toluidine blue solution.
Alkaline phosphatase activity was intense only in proximal tubule and no activity was found in the other parts of the tubules-in the nophron.
Acid phosphatase activity was seen as granular and was most abundant in proximal tubule cells, but the rest of the tubule had little or no activity.
ATPase activity was seen only in brush border of the proximal tubules and reaction was weak in the cell membranes of the rest of the tubule cells.
