Abstract


Histochemical activities of several phosphatases were investigated on the developing liver of sixty-eight cases of human fetuses from 2.3cm C.R length to full term and results obtained were as follows.
1. Alkaline phosphatase:
Formol-calcium or glutaraldehyde fixed sectiones stained with Gomori’s cobalt technique and Burstone´s azo-dye coupling method showed marked enzyme activity, in sinusoidal lining cells, central vein, and -sublobar vein, whereas bile canaliculi, bile-duct and lymphatic vessels showed no activity.
Portal vein branches were generally not stained with rare exceptions of faint coloration although staining was pronounced in capillaries and arterioles of portal area.
Aoy appreciable differences in intensity and distribution of the enzyme were. noted in the liver of different intrauterine month.
2. Adenosine triphosphatase and adenosine monophosphatase:
Formol calcium fixed or non-fixed fresh frozen sections were incubated in Wachstein-Meisel´s medium. Results with both substrates were essentially similar although AMP-ase activity was generally weaker in blood vessels of portal area than those of ATP-ase.
Strong enzyme activities were always demonstrated in bile canaliculi and blood vessels of portal area. Also, lead sulfide was often deposited in sinusoids and central veins. Fresh frozen sections were not as suitable as sections from tissues fixed in cold formol calcium. Frozen sections showed varying degree of issue damage and irregular activity in bile canaliculi. These structures, however, were stained regularly in fixed sections.
3. Non- specific glycerophosphatase:
Formol- calcium fixed frozen sections were incubated at pH 7.2 in a madium containing ?- glycerophosphatase. Moderate or weak activtity of the enzyme was noted in hepatic cell cytoplasm and still weaker reaction was often observed in sinusoid and blood vessels of portal area.
4. Glucose-6-phosphatase:
Fresh unfixed frozen sections were incubated in Wachstein-Meisel’s medium. Very weak activity of the enzymes was observed in the cytoplsm of hepatic cells, the cells of periportal area, however, showed more pronouced activity than those of the central area. Glucose-6-phophatase was assayed by determining inorganic phosphate liberated from glucose-6-phosphate by the liver homogenate. Average activity of sixteen cases examined was 2.0017?0.5588 μ mole phosphate/min/g. wet. Liver tissue of fetus.
No significant differences of the enzymes activity were found in the liver of different intrauterine month up to the end of 9 months.
