Abstract


Stopflow analyses were-performed to gain some insights into the mechanism and site of the diuretic action of probenecid, a uricosuric, during bicarbonate infusion in the dog.
Probenecid inhibited reabsorption of water and sodium along the entire length of the tubule, i.e. proximal, distal as well as intermediate portion. Potassium reabsorption also was depressed and a marked secretory peak in the distal portion was observed.
The reabsorption of bicabonate was also markedly depressed along the entire nephron, and pH of the stopflow samples increased in the distal portion. Secretion of para-amino-hippuric acid was completely abolished by probenecid.
All the observed facts can be best explained on the assumption that probenecid inhibits the carbonic anhydrase in the tubule. Thus it produces diuresis, by increasing urinary excretion of bicarbonate, sodium as accompanying cation, and water bound osmotically.
