Microscopical studies on the testis and some related organ of the vasectomized mouse.

Abstract


In order to investigate the influences of : vasectomy on the testis, epididymis and hypophyses,ry vas deferens of both side of the adult mice were ligated and cut by varing time interval from one to twleveweeks after the vasectomy animals were killed and tissues of the organs were fixed for overnight ´in cold folmol- calcium or in glutaraldehyde and were studied histologically and histochernically. Results obtained were summerized.as follows:
1. Body weight and weight of testes of the control and vacectomized mice were compared at the various time interval after vasectomy and no significant differences were found between the two groups of the animals.
2. On histological examination, no detectable changes were observed in seminiferous epithelium and in interstitium after vasectomy, although diameters" of the tubule and the lumen were significantly increased in the vasectomy group.
3. Many spermatogonia were labelled 3,hrs. after the injection of l-isthymidine into the tail vein and no significant difference in the labelling index was found between the groups of animals.
4. Alkaline phosphatase activity was intense and well localized on the -basement membrane surrounding the seminiferous tubule b any etectable differences in the distribution and intensity_.b groups of animals. without showing etween the two
5. Marked activity of acid phosphatase was noted in the Leydig cells and the Sertolicells.´ The activity in ther´:Sertoli cells appeared to be` most prominent at about the time when the sperm and residual bodies were pre-en in the` lumen of the seminiferous tubules. in the vasectomized group, Tthe enzyme activities appeared to, be slightly more pronounced than those of the control.
6. Sudan Black B stained the granules of the Leydig cells and residual bodies of the seminiferous tubules, in which no significant differences were observed between the control and vaseetomized animals.
7. Significant changes were noted in the epididymis of the mice from two weeks after vasectomy. Lumina of the ductus epididymis were dilated ´ and the sperm were packed within the lumen. These changes were gradually enhanced as the time interval after vasectomy was prolonged. In- some of the extreme cases partially damaged tubules. round ´ cell infiltration and the fibrotic. changes-were also found in the epididymis of prolonged experimental group.
8. At the luminal´and basal surfaces of the ductus epididymis, alkaline phosphase activity was intense and no significant differenences the activity were noted between the control and the , vasectomized mice.
9. Granules reactive to the acidphosphtase were numerous in the epithelium of -ductus epididymis of the both :group and these granules =slightly increased from 6 weeks after vasectomy.
10. Sudan Black B stained granules a certain cell (sudenophilicin the interstitiuni of the epididymis, these sudanophilic cells were few andramdom in`control animal, ´however, they were, abundant and appeased to be arrangel mainly near the base of the epididymal ducts.
11. Hypophyses: No significant differences were noted -´ i histological section between the control and vasectomized animals.
From these results it may be concluded that. the vasectomy_ of does not cause any noticeable histological and histochemical changes_testes and hypophyses ini-the.epidipymis, however, some of she significant changes including fibrosis noted from 6 weeks after vasectomy may,-eause partial or total block of sperm passage.
