Abstract


It was reported by Hammel et al. (1964) that isolated nuclear preparation from pigeon red Woad cells synthesizes hemoglobin in the presence of glucose, as evidenced by the incorporation of 14C-labeled amino acids into the saline extractable globin. In the present studies their findings were confirmed with the nuclear preparation from. chick erythrocytes, and the effect of NAD on this process has been ;investigated since an enzyme preparation from mammalian nuclei was reported by Hayaishi et al. (1967) to catalyze the polymerization of NAD into a polymer with repeating ADP-ribose units (poly ADP-ribose) with the simultaneous release of nicotinamide.
Sucrose-washed nuclear preparations from saponin-hemolyzed mature erythrocytes were incubated with "C-leucine in the presence of NAD, nicotinamide, poly ADP-ribose, or heroin. The incorporation of the label to the saline-extracted protein (hemoglobin) was depressed when NAD - was added to the incubation mixture. The inhibiiion Eby NAD was roughly -Proportional-to the concentration of NAD 0;05-1.0 mM), and released by nicotinamide.
in confirmation of Hammel and Bessmans´ findings (1965), hemin stimulated the incorporation of the label into the protein, and this stimulation was further enhanced when NAD was added together. Time -course study of leucin incorporation showed that the inhibition of NAD was apparent only after the period of 30 minutes.
