Abstract


The present experiment was performed to find out the cause of anemia which appears in the experimental hypersplenism. The serum and splenic emulsion from the normal rabbits and those from the animals with an experimental hypersplenism were administered to two different group of normal rabbits, and the changes of peripheral erythrocyte count were checked; the hypersplenism was produced by the prolonged administration of methylcellulose. At the same time, the activities of reticuloendothelial system in normal and hypersplenic rabbits were examined by means of radioactive tracer method for comparison, and the following results were obtained.
1) Intraperitoneal injection of diluted hypersplenic rabbit serum caused marked decrease in number of circulating erythrocytes, whereas that of normal serum caused no significant changes.
2) Intraperitoneal injection of normal or hypersplenic spleen emulsion caused similar decrease of circulation erythrocytes.
3) In the hypersplenic rabbits, an increased reticuloendothelial function was demonstrated by the accelerated disappearance rate of the reticuloendothelial blocking agent (Silver protein^(-131)I).
4) Distribution of reticuloendothelial blocking agent in various organs appeared to be altered in the hypersplenic rabbits; the phagocytic activities of the lung and liver were increased markedly, whereas those of spleen and bone marrow were decreased slightly.
5) The experimental evidences suggest that the certain humoral factor which may control the productio of erythropoiesis does exist, morrower, it seems that the hypersplenic serum contains greater amount of such humoral factor than the normal serum, and it is most likely not due to the increased secretion of the unit tissue of the spleen but to the relative increase of spleen mass in hypersplenism. Not only the increased destruction of the red blood cells due to the augmented phagocytosis but also the relative incease of humoral factor seem to be the cause of anemia associated with the experimental hypersplenism.
