Abstract


To test the possibility that malonic acid serves itself, as a precursor of citrate excreted in the urine during malonate-induced citraturia, 14C-labeled malonate was infused into a renal artery, while the other kidney received the same amount of unlabelled malonate, and urines from both kidneys were collected separately. Citrate from urine samples were separated by column and paper chromatography and their radioactivity, was determined by liquid scintillation counting. Urine samples from labeled malonate side contained more radioactive citrate than the contra lateral side, and the estimated tubular synthesis and secretion fraction (ETSF) reached 10-16 percent during labeled malonate infusion.
This study clearly shows that, in addition to the decreased reabsorption of filtered citrate, a significant portion of citrate excreted in the urine during malonate infusion is derived from malonate, itself, and that malonate can serve as a precursor off urinary citrate. The pathway of malonate metabolism is yet to be elucidated, but the activation to malonyl coenzyme A and conversion to acetyl coenzyme A would result in direct link to the tricarboxylic acid cycle.
