Abstract


Kallidin, a decapeptide derived from an e;,?globulin, is known to induce severe hypotension and antidiuresis when given intravenously, but it produces diuresis when it is applied directly into a renal artery. Tn this studies, the intrarenal action of kallidin was further explored, especially in reference to the intrarenal distribution of blood flow and osmotic gradient in the medulla produced by the counter?current multiplier system.
When infused directly. into a renal artery in doses of O. 1pg/kg/min. , kallidin elicited a prompt and brisk diuresis confined only to the infused side, with concomitant rises in osmolar clearance and sodium excretion as well as a marked increase in free water clearance. Total renal plasma flow and the medullary plasmm flow increased markedly, while glomerular filtration rate remained unchanged. Corticomedullary concentration gradient of sodium also markedly lowered in the infused kidney.
These changes were also observed during mannitol diuresis, though they are markedly weaker. During maximal diuresis by ethacrynic acid, the kallidin effects were not apparent.
From these observations it is concluded that the increment of medullary flow, effecting washout of the hyperosmolarity created by the countercurrent system, contributes to the diuresis induced by kallidin, which presumably inhibits reabsorption of sodium mainly in the proximal portion.
