Abstract


Histochemical study on alkaline phosphatase has been carried out on the distribution and intensity of the enzyme in the gastro -intestinal canal of the vertebrate applying Burstone´s method using Naphthol AS?TR phosphate as substrate and Fast-red violet LB salt as coupler. Incubation time was 20 minutes at room temperature. And results obtained were as follows.
1. No activity was found in the mucosal surface of the stomach and large, intestine of all animal species examined except in the large intestine of the dog in which mild reaction was noted at the mucosal and glandular cell surfaces.
2. Strong activity was noted at the free border of the villus mucosa and the intest:na:l gland near the opening of the crypt and these cells showed intracytoplasmic activity in the supranuclear region of the cells, corresponding to the Golgi area of the cells. Degree of the enzyme activity in both sites seemed to be roughly proportional to each other. Activity and distribution of the enzyme appeared to be different according to the animal species, the intestinal level, and portions of a villus. It was, generally, strongest in the proximal half of the small intestine in the rat, mouse and dog, weakest in the proximal half of the starved frog intestine. In most animals, mucosal cells in the distal half of the small intestine showed reduced activity of the enzyme which disappeared, often, in the terminal ileum.
3. Mesenchymal enzyme activity noted in the various levels of gastro-intestinal tracts of the animals showed differences in its intensity and distribution according to the animal species and level of the canal wall. Although, most animal shotivcd intense activity in blood vessel and some cellular components, mostly in leucocytes. It was, generally, strongest in she stro-intestinal canal wall of the tortoise, in which muscularis thucosae and outer loalitudinal muscle presented prominent activity besides in the blood vessels. There was marked linear activity just outside the lamina propria of the carp intestinal wall.
