Abstract


The effect of 2, 4 dinitrophenol, an uncoupling agent of oxidative phosphorylation, on the renal function of the tortoise was investigated in this study. The drug was administered into a hindleg vein, which leads to the renal portal circulation, and the other side served as control. A small dose of DNP, 8μg/kg/min, produced slight increases in urine flow and sodium excretion, as well as the creatinine excretion. A large dose, 200μg/kg/min elicited a marked decreases in urine flow and excretion of electrolytes with highly correlated decrease in creatinine excretion; indicating that the renal effects of DNP, are consequences of changes in glomerular function, though it is not clearly defined. Direct effect on tubular transport of sodium is not likely. On the other hand, the secretion of PAH was completely depressed by the agent. It is postulated that the sodium transport in the renal tubules of tortoise, as in mammalian kidney, is coupled to the oxidation prior to the phosphorylation in the electron transport system.
