Abstract


The excretion of various sulfonphthalein dyes such as phenol red (PSP), bromphenol blue (BPB) and bromcresol green (BCG) in both urine and bile was studied in 44 anesthetized dogs. A single dose of a dye (20mg/Kg) was given intravenously at zero time following which urine and bile samples were obtained every 30 min. for 3 hours. Normally, 75% of PSP given was found to be excreted in urine in 3 hours while only 2.7% appeared in bile, indicating a faster rats of PSP secretion in the kidney than in the liver. The secretion of PSP and BCG was, however, much faster in the liver than in the kidney. When Diodrast (350mg) was administered intravenously one hour after the injection of a dye, the excretions of the dye in both urine and bile were significantly reduced in all dyes studied. A similar reduction in the dye excretion was also observed when the experiment was carried out in hypothermic dogs(28 to 30℃) without diodrast. When the corresponding clearance values and the urine/plasma or bile/plasma c9ncentration ratios were calculated for each dye, it was found that these values were generally lowered either by the Diodrast administration or in hypothermia, suggesting that these dyes are secreted actively by a common transport system in both the kidney and the liver. However, a detailed analysis indicated that there are quantitative differences in handling these dyes by the two organs.
As the second step, the secretion of each dye was exclusively studied in the liver in bilaterally nephrectomized dogs. In this series, bromsiilphalein (BSP), a phthalein dye, was also studied along with the three sulfonphthalein dyes. The dye excretion was greatest in BSP and decreased in the order given; BCG, BPB and PSP. This result strongly indicates that BSP, though not a sulfonphthalein dye, is probably secreted by the liver through the same transport system as sulfonphthalein dyes.
