Abstract


The histological findings of the nasal mucous membrane and tissue mast cells in female rats, during the normal cycle, pregnancy and parturition periods, were studied. Also the influences of the artificial changes of the normal cycle, and the hormonal effects on the nasal mucous membrane and tissue mast cells were observed.
Specimens were taken from the posterior part of the nose (just front of the orbit) by total vertical dissection so as to expose the whole nasal cavities including bilateral conchae and septum in one preparation.
Mammary gland and uterus of the same animals were also examined histologically for comparative studies with the nose.
Preparations were stained with 0.1% and 0.5% toluidine blue solution, and hematoxylin-eosin.
1. The nasal mucous membrane and its mast cells show definite periodic changes during the normal sexual cycle, and these changes are almost parallel to the changes of the mammary gland and uterus of the same stage.
2. Tissue mast cells in the nasal mucous membrane are very few during the phases of proestrus and estrus, but markedly increase during the phase of metestrus, and again decrease in the phase of diestrus.
3. During the periods of proestrus, estrus and diestrus, shapes of mast cells in the nasal mucous membrane are mostly dispersed type, and dark stained with some vacuolations or degranulation, but relatively oval shaped masts cells appeared in metestrus stage
4. The nasal mucous membrane show marked increase of gland tissue with secretory appearance during the period of pregnancy. Tissue mast cells are not observed at all during the early part of the pregnancy, then shows slight increase in the later part, and again decrease in 24-48 hours after parturition.
5. The experiment to maintain the estrus 1cr 4 days with intradermal injections of estrogen shows very similar histological findings i n the nasal mucous membrane as estrus, but the number of mast cells in the mucus decreases markedly.
6. The experiment to disrupt the cycle for 2 weeks with intradermal injections of progesterone shows the very similar pictures of the metestrus, but the number of mast cells in the nasal mucous membrane decreases more markedly than in the normal metestrus.
7. The nasal mucous membrane of the castrated female rats show moderate atrophy of gland tissue, and the numbers of tissue mast cells are few.
8. Administration of estrogen or progesterone to castrated female rats produced edematous changes in the nasal mucous membranes and atrophy of gland tissues. These histological changes appeared to be exaggerated when the dose of the hormone injected is large or the duration of treatment is prolonged
9. Administration of estrogen or progesterone to castrated female rats exerted marked decrease of tissue mast cells in the nasal mucous membrane, mammary glands, and uterus: No significant relationship is observed between the degree of mast cells decrease and dose or duration of the hormonal administration.
10. Tissue mast cells in the nasal mucous membrane during the hormonal administration are mostly of diffuse dispersed type.
