Abstract


In order to investigate acetate-C14 metabolism in bone, 120 male mice, weighing about 20 gm, were devided into 6 groups.
Group 1, control (for in vivo experiment)
Group 2, 1.5mg of cortisone acetate injected
Group 3, 0.3mg, of DOCA injected
Group 4, control (for in vitro experiment)
Group 5, to incubate with 1. 5 mg of cortisone acetate
Group 6, to incubate with 0.3 mg of DOCA
Bone, fracture was performed on left femur. Two days after bone fracture mice were sacrificed and both femurs were isolated.
Each femur was weighed, pulverized in mortar and homogenized with incubation- media containing acetate-C14 in glass homogenizer. After incubation (38°C, 2 hours) in incubation flask, CO2 production rate, specific activity of CO2, content and specific activity of cholesterol; amount and specific activity of saponified fraction.
From the data, thus obtained, the author concluded that;
1) CO2 production rate and its specific activity were increased significantly by bone fracture and cortisone
2) Content of cholesterol, saponified fraction and their specific activities were decreased by cortisone
3) DOCA did not effect significantly on metabolism of acetate
4) The effects of cortisone, were similar both in vivo and in vitro
