Abstract


The mechanism involved in the reduced bactericidal power of brucella-immune sera against brucella organisms has been investigated with the following results.
1. The brucellacidal activity of brucella-immune sera was decreased, regardless of the kind of brucella antigen employed for the production of the sera, compared with that of normal serum.
2. Physiological saline solution was found highly toxic to brucella organisms. This toxicity helped differentiate effectively the brecellacidal action of each serum.
3. Addition of complement to five-fold serial dilutions of brucella-immune serum did not enhance the brucellacidal activity of the serum, suggesting that the reduced brucellacidal activity was due to neither the lack of complement nor excess of antibody; the reduced brucellacidal power could not be interpreted by the prozone inhibition phenomenon theory.
4. The blocking antibody, which may be developed in the later stage of immunization, was not detected in the brucella-immune sera tested, suggesting that the agglutinationblocking antibody could not be the cause of the reduction of brucellacidal activity.
5. Absorption of antibody could cause a little recovery of the brucellacidal power of immune sera, suggesting that immune bodies were interfering with the brucellacidal action of serum.
6. The mechanism for the reduction of brucellacidal power in brucella-immune sera was discussed and the following hypothesis was proposed Brucella-immune bodies may play a role of protecting brucella orgsnisms from the brucellacidal action of natural antibody and complement inasmuch as the cellular immunity is mainly concerned with the host resistance in brucellosis, the circulating antibodies´ reaction with brucella organisms in vitro may be said to form a kind of buffer zone between the brucellacidal agents and the organism.
