Abstract


Many sexually matured mice weighing 20g on an average were used in this study. The male mice were divided into 3 groups: 1) normal control, 2) castrated and 3) castrated prior to administration of testosterone propionate.
The female ones were also divided into 4 groups: 1) diestrus, 2) estrus, 3) castrated and 4) castrated before administration of estradiol benzoate.
The sexual hormones were injected subcutaneously into the castrated animals twice at an interval of over 24 hours in the amount of 2μg/g body weight. The tritiated thymidine of 0.8μc/g body weight was administered intravenously to all the animals 7 hours prior to sacrifice of the animals.
Pieces of the masseter, deep back and rectus femoris muscles were fixed in Bouin´s fluid and made into paraffin sections 2-3μ thick. The slides were prepared for autoradiography by the stripping film method and stained with Harris´ hematoxylin.
The number of labeled cell nuclei in the muscles was counted among 1000 randomly observed cell nuclei and the ratio, to wit the radioactive index, stands for estimating the value of DNA synthesis or the cell renewal rate.
The results are summarized as follows:.
1. When the gonads were removed from the mice, the radioactive indices of the muscle cells were decreased significantly as compared with those of the normal group; whereas in the event of the hormonal supplement, the indices were increased up to normal level or above that.
2. In the female radioactive indices there was a more significant increase in muscles in diestrus than those in estrus.
3. In both sexes of the normal mice, the radioactive indices of the rectus femoris
were about two tunes higher than those of the back and masseter muscles, while those of the back muscle were reduced most evidently after castration.
4. It comes to a conclusion that the sexual hormones injected supply the hormonal deficiency in the castrated animals and further activate DNA synthesis or the cell renewal of the mice muscle to normal level or upwards.
