Abstract


In order to study the effects of pancreatic duct ligation on gastroduodenal mucus secreting cells in rabbits, this work was performed. Healthy adult male rabbits weighing about 2kg body weight were divided to the normal and pancreatic duct ligation groups. Ligation of ire pancreatic duct was done aseptically under sodium nembutal anaesthesia, and animals were kept under room temperature. Duct ligated animals were successively sacrificed by ether inspiration at 3 days, 1, 2 and 3 weeks after operation. Specimens of gastric and duondenal mucosa were immediately fixed in 10% buffered neutral formality solution, embedded in paraffin wax, sectioned by six micra-thickness, and stained chiefly with alcian blue(pH 2.5)-PAS(periodic acid Schiff reaction) and colloidal iron-PAS, with additional stainings of alcian blue(pH 2.5), 0.02% azur A(pH 1. 5,3. 0,4. 5) or alcian blue(pH 2.5)-K.ernecht-rot to ascertain comparison with the reaction of chief stains.
Observed results are as follow:
1. In the gastric mucosa of rabbits, the surface epithelium reveals alcianoph-ilia, and purple red or deep red massive mucous substance in the supranuclear cytoplasm of them by alcian blue (pH 2.5)-PAS reaction, i.e., bluish properties predominate in the superficial convolution and show apparently the redder in the foveolar walls. The mucous neck cells show red and secrete an apparently neutral mucosubstance which differ from the surface epithelium. The chief cells in the upper portion of gastric glands reveal alcianophilia and show weak blue tint. The parietal cells in the upper and middle portion of the glands show very weak red in the luminal cytoplasm.
2. Gastric foveolae are slightly enlarged at 3 days after the duct ligation. Blue tint in the chief cells and the red in the parietal cells increase deep in the glands. At one or two weeks after the ligation, atrophy of mucosa, especially of foveolar epithelium is observed. Columnar epithelial cells show reddish tint, and the parietal and chief cells which have s-taina´ele mucosubstance in the supranuclear portion of their cytoplasm are found in the middle and lower portions of the glands. In this stage, decrease of alcianophilia and. relative increase of PAS positive reaction is evident.
At 3 weeks after the ligation, morphological return to control group, except increase of red. density of the parietal cells in pyloric portion is noticed.
3. In the erosed mucosa or the mucosa near the ulceration, interruption of purple mucus coating over the surface epithelium and decrease of alcianophilia of the chief cells are accompanied with hyperplasia and ballooning of the parietal cell cytoplasm occur and this phenomena indicate morphological destruction of the protective balance of the mucosubstances of gastric rnucosa under pancreatic duct ligation.
4. In the duodenal mucosa of normal rabbit, the Brunner´s gland cells reveal alcianophilia in luminar cytoplasm. the goblet cells show various tints from blue to violet according to level in villi and, in general, alcianophilia predominates in the cells located at superfical or upper portion of villi.
5. Exfoliation of the surface epithelium, atrophy of the mucosa, erosion or ulceration occur in the duodenum, especially, near the. pancreatic duct orifice, after pancreatic duct ligation. Alcianophilia of the Brunner´s gland cells slightly decreases at 3 days, but increases at one or two weeks after the ligation. Increase of number of the goblet cells in the duodenal epithelium and their staining variability
from blue to violet indicate hyperactivity of the cells under pathologic condition produced by pancreatic duct ligation.
At three weeks after the ligation, alcianophilia of the Brunner´s gland cell and the goblet cells returns nearly to normal condition, but reveals somewhat strong reaction rather than those of the normal ones.
6. In or near the duodenal mucosa with erosion or ulceration, interruption of the blue or purple mucous coating covering viii and decrease of alcianophilia of the Brunner´s gland cells and goblet cells are noticed. This figures suggest the destruction of the protective role of mucous substances in the duodenum.
