Abstract


Colonial variants of Pseudomonas aeruginosa are frequently induced spontaneously and many investigators isolated various mutants according to their markers. However, most of workers agreed with the findings that those mutants were not different from original parent forms substantially on both morphological and biological characters other than phage sensitivity and conflicting opinions of pyocins production.
In the present report, the authors have studied the colony dissociation in Pseudomonas aeruginosa from old broth culture with isolated stock strains and pyocin typing indicators, and found that about half of the tested strains dissociated mutant colonies on agar plates. From each date 4 colonies were picked up to compare the properties with original colonies.
Although no detectable differences were observed in cell morphology microscopically, there were marked differences in colonial morphology of mutants such as; small size (ranging from 1/3 to 1/10 in diameter) dry and winkled surface, sharp edge, somewhat non-pigmented appearance, and R form character of difficulty to disperse in saline and predominant pellicle formation in broth. The motility of some variants was weakened or lost and the growth rate in tryti case soy broth was much delayed than original forms.
As far as biological characters are concerned, no differences were demonstrated in pyocyanin production, oxidase activity, and sensitivity to streptomycin, polymy in B and colistin, although some differences were noted in streptomycin sensitivity in some strains of mutant.
Pyocin production of the variants was not different from that of original forms on the blood agar, although some differences in pyocin patterns were exhibited in synthetic media.
