Abstract


It is apparent that endorcrine glands give some influences to allergic changes. Since "stress" reaction has been postulated by Seyle, the endocrinologic studies of steroid hormones have become as an essential subject to clarify the mechanism of allergic phenomenon.
Professor Yun described the marked diminution of anaphylactic shock in the adrenocortical substance, internin, administrated animals, prior to the inhibition of allergic changes in ACTH or Cortisone treated patients is reported. Meanwhile, Sakai recognized the aggravation of allergic changes in the oophorectomized guinea pigs.
The influence of testis on allergic pneumonitis has not been determined. It, therefore, seems of, interest to study on the effect of testis upon the allergic changes- ´in the lung.
Material and MethodNormal rabbits weighing around 2kg were selccted
and divided into 5 groups for this experiment. Group 1: Rabbit sensitized and reinjected, served as control.
Group 2: Rabbits sensitizd and reinjected immediately after orchiectomy.
Group 3: Rabbits sensitized and reinjected, 3 months following orchiectomy.
Group 4: Rabbits sensitized and reinjected,3 weeks following transplantation of testis after orchiectomy done 3 months prior.
Group 5: Rabbits sensitized and reinjected, treated with testosterone following orchiectomy done 3 months prior.
Each of sensitized rabbits was injected lcc of normal horse serum per kg of body weight into the subcutaneous tissue daily for 4 days and the reinjection, O.Icc of normal horse serum per kg of body weight, was given into the lung parenchyma 15 days later.
Testicular hormone used in the experiment was 10 mgm of testosterone propionate per kg of body weight which is distributed by the Pacific States Laboratories Inc., U. S. A.
All rabbits were sacrificed at the end of 3, 6, 12, 24, 48, 72 hours and 1, 2 weeks respectively after the
final injection. The specimens were prepared by H. & E. and Van-Gieson stains.
Results
1. In group 1, macroscopically, edema, congestion and hemorrhage were present. Microscopically, cloudy swelling, fibrinoid degeneration, hemorrhage, infarction or thrombosis, inflammatory exudates and cells, neutrophiles, lymphocytes, eosinophiles, monocytes and plasma cells were observed. Desquamated bronc hiolar epithelium were also noted.
These changes appeared in 3-6 hours were reached their maximum at 24-48 hours, however, gradually subsided in 1-2 weeks.
2. In group 2, somewhat more aggravated pictures of the above montioned changes were observed in 3 hours compared with the control group, group 1.
3. In group 3, much fewer inflammatory cells, especially, eosinophiles and aggravation of the de-generative changes compared with the control group were observed.were similar in its intensity as in those of control group.
Conclusions
Experimental studies using rabbits on the influences of testis upon the allergic changes in the lung are reported.
1. The allergic changes in the lung of orchiectomized rabbits revealed somewhat more gagravated than in those of the non-orchiectomized control rabbits.
2. The allergic changes in the Iung of rabbits 3 months following orchiectomy, were much less than in those of control rabbits.
3. Either in group 4 or in group 5, the over all allergic changes in the Iung were similar ´ in inteusity as in those of control group. In group 4, the degenerative changes were somewhat more aggravated. Whereas in group 5, inflammatory cell infiltration, especially of eosinophiles, was somewhat more conspicuous.
4. By this experiment, it may be concluded that. resection of the testis restrains the allergic changes but administration of testicular hormone (testosterone) enhances the- allergic changes in the rabbit lung.
