Abstract


1. Glutamic-oxaloacetic ´(GOT) and glutamic-pyruvic transarninase (GPT) of dog´s submaxillary gland were partially purified by ammonium sulfate fractionation method, and the effect; of the addition of pyridoxal phosphate and various metal ions on the activity of the enzyme preparation as well as of whole homogenate of salivary gland were studied.
2. Considerable loss of the GOT activity was noted to occur during. the purification. On the other hand, the inactivation of GPT during the purification was not so marked as in GOT.
3. The addition of pyridoxal phosphate (2.5 mM) to whole homogenate prior to proceeding to the purification prevented the loss of GOT during the purification, resulting in the increase in purification degrees. However, the activity of GPT was not so markedly increased by the prior treatment of pyridoxal phosphate. The same situations were also observed to hold with the partially purified enzyme preparations of GOT and GPT. That is, the addition of pyridoxal phosphate (10-3M) caused the almost 5-fold increase in the activity of GOT preparation whereas only 2-fold increase in GPT activity was resulted.
4. From these results, it was suggested that the apocoenzyme combination of GOT found in dog submaxillary gland is not so tight as that of GPT, and is easily dissociable.
5. General enzyme inhibitors such as Ag+, Hg++ and Au++ were also very inhibitory to both transaminases at the concentration of 10--6 to 10-4 M. The activities of both transaminases were also inhibited by the addition of Cu++, Zn++ and Co++. GPT activity was more vulnerable to these ions than GOT activity.
6. Fe++ Fe+++ Mn++ Al+++ Cr+v1 and Mg++ ian-,Wo~ , c enzyme activity in the decreasing order. Of these ions, Fe++ Fe+++, and Al... increased the activities of both transaminases, and the other ions were effective only on GPT activity.
7. The effects, either enhancing or inhibitory, of metal ions on the partially purified enzyme preparation with or without the treatment of pyridoxal phosphate, and on whole homogenate were differed each other. How-ever, in general, the activity of GPT was more susceptible to these ions than GOT activity.
